IL2
(increases)
Evidence: We show that Gab2 was transiently phosphorylated by tyrosine in human mycosis fungoides (MF) tumor T cells upon IL-2 stimulation and that SHP2 as well as Stat5a associated inducibly with Gab2. IL-15, but not IL-4, also induced tyrosine phosphorylation of Gab2, suggesting that the IL-2 receptor beta-chain is important for IL-2-induced Gab2 phosphorylation.

:
Adding back each tyrosine to Fall revealed that ( 1 ) Tyr577 was necessary and sufficient for Shc phosphorylation ; ( 2 ) Tyr577, Tyr612, and Tyr695 were
involved in activation of
SHP-2 , the Raf/ERK cascade, and c-fos transcription ; and ( 3 ) all tyrosines, but particularly Tyr612, Tyr695, Tyr750, and Tyr806, facilitated
STAT5 activation

(MAP Kinase Signaling System...) :
SHP2 regulates IL-2 induced MAPK activation, but not Stat3 or
Stat5 tyrosine phosphorylation, in cutaneous T cell lymphoma cells ... Furthermore, the results indicate that
SHP2 may not be
involved in the activation of Stat3 or
Stat5 in CTCL cells

Paradoxically, cell culture studies also suggest a positive
role of
Shp2 in promoting prolactin stimulated

Furthermore,
PTPN9 WT expression specifically
impairs EGF induced STAT3 and
STAT5 activation, and inhibits the cell growth in soft agar

:
A critical
role for
SHP2 in
STAT5 activation and growth factor mediated proliferation, survival, and differentiation of human CD34+ cells ... Inhibition of
SHP2 expression initially enhanced and later
inhibited STAT5 phosphorylation and reduced expression of the antiapoptotic genes MCL1 and BCLXL ... These results indicate an important
role for
SHP2 in
STAT5 activation and GF-mediated proliferation, survival, and differentiation of human progenitor cells
